\/><\/section>\n<section>\u771f\u6838\u751f\u7269mRNA\u7684\u751f\u547d\u5468\u671f\u5305\u62ec\u51e0\u4e2a\u4e0d\u540c\u7684\u9636\u6bb5\uff1a\u8f6c\u5f55\u3001\u524d\u4f53\u4fe1\u4f7f\u6838\u7cd6\u6838\u9178\uff08pre-mRNA\uff09\u52a0\u5de5\u3001\u6838\u8f93\u51fa\u3001\u7ffb\u8bd1\u548c\u8870\u53d8\u3002\u5728\u6bcf\u4e2a\u9636\u6bb5\uff0cmRNA\u4e0e\u4e00\u7ec4\u7279\u5b9a\u7684RNA\u7ed3\u5408\u86cb\u767d\uff08RBPs\uff09\u76f8\u4e92\u4f5c\u7528\uff0c\u5f62\u6210mRNA-\u86cb\u767d\u8d28\u590d\u5408\u7269\uff08mRNPs\uff09\u3002\u8fd9\u4e9bRBPs\u63a7\u5236mRNA\u7684\u6d3b\u6027\u3001\u5b9a\u4f4d\u548c\u7a33\u5b9a\u6027\uff0c\u5e76\u5f71\u54cd\u5176\u5411\u751f\u547d\u5468\u671f\u4e0b\u4e00\u9636\u6bb5\u7684\u8fc7\u6e21\u3002\u56e0\u6b64\uff0c\u63ed\u793aRBPs\u7684\u9636\u6bb5\u7279\u5f02\u6027\u5e93\u5bf9\u4e8e\u7406\u89e3mRNA\u8c03\u63a7\u81f3\u5173\u91cd\u8981\u3002\u7531\u4e8emRNA\u5728\u5176\u751f\u547d\u5468\u671f\u4e2d\u4e0d\u65ad\u6539\u53d8\u5176\u86cb\u767d\u8d28\u4f34\u4fa3\uff0c\u4f46\u7531\u4e8e\u7f3a\u4e4f\u65f6\u95f4\u5206\u8fa8\u7387\u6570\u636e\uff0c\u4f7f\u7814\u7a76\u8005\u4eec\u5bf9mRNPs\u7684\u7406\u89e3\u53d7\u5230\u9650\u5236\u3002<\/section>\n<section>\u4e3a\u4e86\u89e3\u51b3\u8fd9\u4e00\u95ee\u9898\uff0c\u672c\u6587\u4f5c\u8005\u901a\u8fc7\u5728\u7ec6\u80de\u4e2d\u7528\u5149\u6d3b\u5316\u6838\u7cd6\u6838\u82f7\u8fdb\u884c\u8109\u51b2\u4ee3\u8c22\u6807\u8bb0\u3001UVA\u4ea4\u8054\u3001\u57fa\u4e8epoly(A)\u7684RNA<span class=\"wx_search_keyword_wrap\">\u5206\u79bb\u5bcc\u96c6<\/span>\u4ee5\u53ca\u57fa\u4e8e\u8d28\u8c31\u6cd5\u7684\u86cb\u767d\u8d28\u7ec4\u5b66\u6280\u672f\uff0c\u63d0\u4f9b\u4e86\u65f6\u95f4\u5206\u8fa8\u7684mRNA\u76f8\u4e92\u4f5c\u7528\u7ec4\u6570\u636e\u3002\u4e3a\u4e86\u9274\u5b9a\u5728RNA\u5408\u6210\u540e\u7279\u5b9a\u65f6\u95f4\u70b9\u4e0emRNA\u76f8\u5173\u7684RBP\uff0c\u4f5c\u8005\u9009\u62e9\u4e864sU\u8109\u51b2\u8ffd\u8e2a\u7684\u65f6\u95f4\u5206\u8fa8mRNA\u76f8\u4e92\u4f5c\u7528\u7ec4\u5206\u6790\u3002\u4f5c\u8005\u75284sU\u5bf9HeLa\u7ec6\u80de\u8fdb\u884c10\u5206\u949f\u7684\u4ee3\u8c22\u6807\u8bb0\uff0c\u8fd9\u662f\u6700\u77ed\u7684\u65f6\u95f4\uff0c\u4e3a\u86cb\u767d\u8d28\u7ec4\u5b66\u5206\u6790\u63d0\u4f9b\u4e86\u8db3\u591f\u91cf\u7684\u4ea4\u8054\u6750\u6599\u3002\u6807\u8bb010\u5206\u949f\u540e\uff0c\u6d17\u53bb\u6e38\u79bb\u76844sU\uff0c\u7136\u540e\u5b75\u80b2\u5230\u6240\u9700\u7684\u65f6\u95f4\u70b9\u3002\u4f5c\u8005\u9009\u62e9\u4e86\u4ece0\u52305\u5c0f\u65f6\u768410\u4e2a\u65f6\u95f4\u70b9\uff080\u300115\u300130\u300145\u300160\u300190\u3001120\u3001180\u3001240\u548c300\u5206\u949f\uff09\uff0c\u8fd9\u8986\u76d6\u4e86\u5df2\u62a5\u5bfc\u7684\u4eba\u7c7bmRNA\u534a\u8870\u671f\uff08\u5e73\u57476.9\u5c0f\u65f6\uff1b\u4e2d\u4f4d\u65703.4\u5c0f\u65f6\uff09\u7684\u5927\u90e8\u5206\u9636\u6bb5\u3002\u901a\u8fc7\u5c06\u7ec6\u80de\u66b4\u9732\u4e8e\u51b0\u4e0a\u7684UVA\u7167\u5c04\uff0c\u57284sU\u6807\u8bb0\u7684RNA\u53ca\u5176\u5bc6\u5207\u63a5\u89e6\u7684\u76f8\u5173\u86cb\u767d\u4e4b\u95f4\u8bf1\u5bfc\u5149\u4ea4\u8054\uff0c\u5e76\u7acb\u5373\u6536\u83b7\u7ec6\u80de\u3002\u7ec6\u80de\u88c2\u89e3\u540e\uff0c\u7528<span class=\"wx_search_keyword_wrap\">oligo<\/span>(dT) beads\u6355\u83b7\u542bpoly(A)\u7684mRNA\u5206\u5b50\uff0c\u5e76\u5728\u542b\u67094M\u5c3f\u7d20\u7684\u4e25\u683c\u6761\u4ef6\u4e0b\u6d17\u6da4\uff0c\u4ee5\u7279\u5f02\u6027\u6355\u83b7\u76f4\u63a5\u4e0e4sU\u6807\u8bb0\u7684mRNA\u7ed3\u5408\u7684\u86cb\u767d\u3002\u5229\u7528\u8fd9\u79cd\u65b9\u6cd5\uff0c\u4f5c\u8005\u572810\u4e2a\u65f6\u95f4\u70b9\u4e0a\u5bf9700\u591a\u79cdRNA\u7ed3\u5408\u86cb\u767d\uff08RBP\uff09\u8fdb\u884c\u5b9a\u91cf\uff0c\u5e76\u4e14\u8fd9\u4e9bmRNA\u7ed3\u5408\u7684\u987a\u5e8f\u4e0e\u5df2\u77e5\u7684\u529f\u80fd\u3001\u4e9a\u7ec6\u80de\u4f4d\u7f6e\u548c\u5206\u5b50\u76f8\u4e92\u4f5c\u7528\u975e\u5e38\u4e00\u81f4\u3002<\/section>\n<section><img \/><\/section>\n<section>\u6709\u8da3\u7684\u662f\uff0c\u4f5c\u8005\u8fd8\u89c2\u5bdf\u5230\u4e86\u610f\u60f3\u4e0d\u5230\u7684\u52a8\u6001RBPs\u3002\u4f8b\u5982\uff0c\u8f6c\u5f55\u8f93\u51fa\uff08TREX\uff09\u590d\u5408\u7269\u76f4\u5230\u665a\u671f\u624d\u88ab\u68c0\u6d4b\u5230\uff08\u6700\u5927\u5cf0\u503c\u65f6\u95f4\uff0c67\u201377\u5206\u949f\uff09\uff0c\u8fd9\u8868\u660eTREX\u53ef\u80fd\u662f\u5728mRNA\u51fa\u6838\u540e\u518d\u8fdb\u884c\u8f6c\u5f55\uff0c\u8fd9\u6311\u6218\u4e86\u76ee\u524d\u540c\u65f6\u8f6c\u5f55\u548cmRNA\u6838\u8f93\u51fa\u7684\u89c2\u70b9\u3002\u53e6\u5916\uff0c\u5e94\u6fc0\u9897\u7c92\u86cb\u767d\u666e\u904d\u7ed3\u5408\u8001\u5316\u7684mRNA\uff0c\u8868\u660e\u5728mRNA\u751f\u547d\u5468\u671f\u540e\u671f\u53d1\u6325\u7684\u6f5c\u5728\u4f5c\u7528\u3002<\/section>\n<section><img
